Does rib maple give stronger support to the top and back if the grain is
running vertical, or if it is running horizontal? I know if you are using flamed
wood, it should run to match the back. But, what if it is not flamed, does it
make any difference?

It's harder to bend and weaker if it's run the wrong way, and the ribs may also warp in bending in funny ways, which is more what I'd be worried about. You may also have problems with the grains delaminating when bending with slabbed wood. If you can get the ribs bent nicely, though, then they're probably fine to use.
